Abstract:
Emerging applications of elemental imaging have been found in the biomedical and clinical field. There are several examples of bioimaging using laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS), such as brain, liver and kidney. The spatial distribution of biologically essential elements (Cu, Zn, Fe) and other metals (Pt, Gd) can provide valuable information for understanding the accumulation and metabolism of the elements. In the present work, we have successfully performed bioimaging using LA-ICP-MS in mice liver, kidney, as well as mice whisker after drug administration, demonstrating the potential use of LA-ICP-MS in drug development study and biomonitoring. Cis-platin is the first line anti-cancer drug and it is known to be nephrotoxic. In the first part of this project, the anti-cancer potential of some metal containing complexes were evaluated. Two platinum complexes were chosen and administered to ICR mice by intraperitoneal injection. The distribution of metal in brain, lung, heart, liver, kidney and testis were determined by ICP-MS bulk analysis. Besides, the spatial distribution of platinum for these complexes in formalin fixed-paraffin embedded liver and kidney sections were studied by LA-ICP-MS.
梁士贤于香港大学化学系获得博士学位。现任香港浸会大学分析化学理学硕士课程总监、环境与生物分析国家重点实验室首席研究员、化学分析中心副主任、化学系副教授及香港大学荣誉副教授、博士生导师。兼任香港特区政府卫生署属下的香港中药材标准国际科学委员会成员,创新科技署属下的药剂研究专责小组顾问、检测和认证专责小组顾问、认可委员会(化学分析)委员,主任评审人员等。他是Analytical Chemistry, Water Research, Chemical Communications, Analytica Chimica Acta等10余种国际学术期刊的编委或编审人员。目前主要研究方向包括环境与生物分析化学、中药分析化学、质谱应用于临床诊断等。最近五年承担多项国内、外研究课题, 获得科研经费逾千万。已发表SCI研究论文逾百篇,多部书籍、系列丛书的编委。
