Biomaterials for cell and tissue engineering

Abstract:
The biochemical and biophysical properties of biomaterials have profound effects on cell and tissue functions. We are interested in understanding how the biophysical properties such as the micro/nano structure and stiffness regulate cell behavior and how the bioactive materials can be used to promote tissue regeneration. Here I will discuss two examples: In vitro cell reprogramming and in vivo tissue regeneration. Stem cells are a valuable cell source for tissue engineering, disease modeling and drug screening. A recent discovery in stem cell biology is that differentiated cells can be reprogrammed into induced pluripotent stem cells (iPSCs) and desired cell types. Although the effects of transcriptional factors and chemical compounds have been widely studied, the role of biophysical factors on cell reprogramming is not clear. I will present our findings on how biophysical factors can regulate the epigenetic state and thus the cell memory and reprogramming process, which has important implications in cell conversion into iPSCs and specific cell types. To illustrate the important role of biomaterials in tissue regeneration and remodeling in vivo, I will use blood vessel regeneration as an example to demonstrate an evolution from in vitro tissue engineering to in situ tissue engineering approach. In this approach, endogenous stem cells are recruited by the use of bioactive scaffolds to promote tissue regeneration.
个人简介:
李松,理学博士,美国加州大学伯克利及洛杉矶分校生物工程系教授,上海交通大学讲席教授,重庆大学荣誉教授。在北京大学获学士及硕士学位;在美国加州大学圣地亚哥分校生物工程系师从钱煦教授获博士学位。李松教授主要从事干细胞工程、血管组织工程、血管修复、微纳米技术和生物材料等方面的研究。近五年来,发现了一类新的血管干细胞及其在血管重组中的作用。另一方面,首次发现了生物材料和生物物理因素对细胞重编程的促进作用。担任4个国际杂志编辑,为60余个学术期刊审稿,为20余家国际学术机构提供各类学术建议和评估。选为美国医学研究所、生物工程和生物医学工程学会以及国际医学和生物工程学院的高级会员,并担任2016年全美生物医学工程全国大会主席。
